Title

Validation characteristics for the measurement of Amnesic Shellfish Poisoning toxin.

Objectives

Domoic acid, the toxin responsible for Amnesic Shellfish Poisoning (ASP) has been recorded in commercially important shellfish including scallops, mussels, oysters, clams and cockles. The procedure for the extraction of toxin from shellfish tissue prior to analysis involves the use of 50% aqueous methanol followed by measurement using HPLC. For accurate work such as the preparation of Certified Reference Material, exhaustive extraction from the shellfish tissue is recommended, however regulatory laboratories, under pressure to provide results, generally use a single dispersive extraction.

This project will investigate the extraction efficiency of domoic acid from a range of shellfish tissues including the adductor muscle, hepatopancreas, gonad and tissue remainder from the king scallop, Pecten maximus; and from total tissue of the mussel, Mytilus edulis, the cockle, Cerastoderma edule, the native oyster, Ostrea edulis; the Pacific oyster, Crassostrea gigas and the Manila clam, Ruditapes philippinarum, all commercially important species in Europe. Extraction efficiency will be investigated over a range of concentrations, both above and below the 20µg/g concentration identified in EU Directive 2002/226/EC.
